It is supplied as a powder which must be reconstituted into a 100ml
working solution.
Description:
Metaphase Arresting Solution is a mixture of colchicine and vinblastine
sulphate dissolved in PBS. Metaphase Arresting Solution is a gentle
alternative to commonly used colchicines/colcemid preparations and
is less toxic to cells because the concentration of colchicine is
reduced while the extra blocking activity of vinblastine sulphate.
How
it works:
Both colchicine and vinblastine sulphate inhibit the formation of
mitotic spindles and are used to increase the percentage of metaphase
cells for chromosome analysis. It is the additional presence of
vinblastine sulphate that makes Metaphase Arresting Solution effective
with bone marrows, difficult tumor samples, and problematic tissues
with low mitotic indexes, where colchicine/colcemid alone
appears to be ineffective. Vinblastine sulphate is a gentler metaphase
block.
How
to use it:
Metaphase Arresting Solution is supplied as a powder and when reconstituted
as specified on the package insert becomes a 100ml working solution.
Metaphase Arresting Solution is simply substituted for standard
colchicines/colcemid preparations used in standard harvesting protocols.
This means that about 60-100 microliters of Metaphase Arresting
Solution is added per 10ml of cell culture.
Benefit
to Lab:
The use of Metaphase Arresting Solution is effective in increasing
the mitotic indexes of bone marrows, difficult tumor samples, and
problematic tissues where colchicine/colcemid alone appers
to be ineffective.
